Hi! do i still need to use a DI box if I'm plugging the bass guitar into a ROLAND cube 30 bass amp, then use its rec out into my audio interface? I'm using Yamaha MG16XU?

Depends on whether you're using the mic input or the line input on the mixer. If you're using the line input then the Rec out should be a good level and impedance match and no DI box necessary. If you're going into the mic input then you should still use the box. If you have hum problems due to ground loops then definitely use the box with the 'ground lift' switch on and go into the balanced mic in on the mixer.

I'm plugging it to a MIC/LINE input on my audio interface, then below it theres two buttons PAD 26db & HPF 80Hz, it has a gain knob (+20/-6) up to (+64/+38). Then there's also a compressor knob, to be more precise you can refer to the below image link to see the unit and would love to hear your advice which input is best to use for recording from a REC/HEADPHONE output of my guitar & bass amplifiers.
