Should I uninstall V4 before attempting to install V5?

As Mario mentioned, you don't have to, but if you do keep ACID Pro 4.0 installed and eventually uninstall it, your file associations might be lost. (It's quickly fixed by uninstalling and then reinstalling ACID Pro 5.0 or by doing a repair.)

I would suggest an uninstall and re-install. In my case though, I have v4.0 and 5.0 set up in different folders. They do not interfere with one another since I have even set up different shared folders for each version. If you have no plans to use v4.0 any further, clear it off your comp first. It's easier to manage orphaned files this way.
