Is it possible to encode a 5.1 AC-3 file with a standard Windows sound card and drivers?

You can encode it yes.

What you won't be able to do is listen to the surround panning to preview what it is going to sound like on a 5.1 system.

Yes, I've done it many times on a laptop. The bummer is you have to burn, waste if you will, a DVD to check your work. However, once you get your timing down you won't waste that many. In fact if you are panning peak sounds you don't have to waste any.
